Summary

This project was designed to assist the Australian Seafood CRC identify research gaps and opportunities and ultimately provide the necessary background information and rationale for a potential supplementary bid to DIISR for funding of a new program of health related research.

The Food Standards Australia New Zealand (FSANZ) website and websites for the Japanese Ministry of Health, Labour, and Welfare and the US Food and Drug Administration were the major sources of information regarding health claims for Australia, Japan and the US. Japan and the US were included in this report as they are two key export markets for the Australian Seafood industry. Examples of seafoods, using specific seafoods of interest to the Seafood CRC were modeled for nutrition content and general level health claims. A brief review of the potential mechanisms of some of the observed health effects of fish and omega-3 fatty acids was undertaken.

The research described in this report found that one of the important mechanisms for the observed widespread effect of seafoods on a range of health outcomes (such as coronary heart disease, cancer, arthritis, dementia) is the dampening of the inflammatory response by the 2 key bioactives in seafood, namely the long chain omega-3 fatty acids eicosapentaenoic (EPA) and docosahexaenoic acids (DHA). However there are other bioactives in fish worth exploring further. Several research gaps were identified as part of this project and these are outlined in the report.

Potential collaborators or partners beyond the current CRC members and affiliates were identified and have been included in the body of the report, as are some early suggestions for alternative funding and funding models.

Evaluating the Southern Bluefin Tuna cell lines as a platform for testing the effectiveness of antioxidants in preserving flesh quality

Need

The vast majority of the Southern Bluefin Tuna (SBT) farmed in South Australia is sold fresh for sashimi production in Japan. Premium prices are obtained for sashimi grade tuna with a high fat content. Unfortunately, fatty fish tissues with their high polyunsaturated fatty acid (PUFA) content are prone to oxidation. This results in poor flesh quality, reduced shelf life and significant trimming of the tuna carcass at the market.

FRDC Project No. 2004/209 investigated the impact of dietary supplements on the flesh quality of farmed SBT. Specifically, it investigated the impact of the dietary antioxidants vitamins C and E and the mineral selenium. Project No. 2004/209 showed that these supplements can extend the shelf life of muscle samples taken from farmed SBT fed diets containing higher than normal concentrations of the antioxidants.

Although Project No. 2004/209 has been very successful, performing diet trials with live SBT is expensive and logistically difficult. It is also time-consuming and the statistical power of the experiments is limited by the fact that it is economically unfeasible to have multiple replicate sea-cages for each dietary treatment. As a result of these constraints, experiments must be repeated over several years to obtain statistically valid results.

Recently, we produced the world’s first SBT cell line and to our knowledge the first cell line for any tuna species. The cell line has the potential to reduce the number of experiments with live SBT and to greatly increase the number of dietary supplements that can be screened in a given period of time. With the cell line, it takes only one week to screen 24 different supplements with 3-4 replicates of each. This compares with years for similar experiments with live SBT.

This project will evaluate the cell line as a platform for testing dietary antioxidant supplements for SBT.

Objectives

1. To adapt existing assays for antioxidant potency from mammalian cell culture systems for use with primary fish cell cultures and immortal fish cell lines.
2. To use the adapted assays with primary fish cell cultures and immortal fish cell lines to screen a broad range of concentrations and combinations of traditional and novel antioxidants for their ability to inhibit lipid oxidation
3. To rank the antioxidants with respect to their antioxidant potency relative to vitamin E, the ‘industry standard’ antioxidant
4. To quantify how the antioxidant concentrations that are effective in primary fish cell cultures and immortal fish cell lines relate to the antioxidant concentrations that need to be added to aquafeeds
5. To undertake a cost benefit analysis of the various antioxidants and antioxidant combinations with respect to their eventual incorporation into feeds
6. To investigate the impact of dietary antioxidants on cellular antioxidants and antioxidant enzymes in primary fish cell cultures and immortal fish cell lines
7. To recommend the best concentrations and combinations of antioxidants to be added to southern bluefin tuna and yellowtail kingfish feeds to maximize fish health and flesh quality

1. To develop sensitive tests using polymerase chain reaction (PCR) to detect a range of viruses of direct significance to human health directly from shellfish. The range would include hepatitis A virus, Norwalk virus, Astrovirus, and Adenovirus.
2. Assessment of coliphage concentrations, and indicator microorganisms as reliable tools for assessment of faecal contamination (Clostridium spp., Bacteroides spp. coliforms) in oyster tissues, and harvest water.
3. Assessment of faecal origin and contamination of oysters and harvest water by Gas-Chromatography-Mass Spectrometry analysis for faecal steroid profiles.
4. To adapt developed biochemical markers in blue mussel (M.edulis) to oysters as indicators of stress from adverse environmental conditions of microbial and chemical origin.
5. Combination and correlation of the above analytical techniques to provide a comprehensive assessment of the extent of contamination of oysters and their harvest waters by pathogens of faecal origin. These techniques would be more reliable than current faecal indicator techniques and allow accurate determination of closure safety zones.
6. Identification and validation of reliable, cost effective monitoring tools for quality assurance of farmed oysters, leases and closure safety zones applicable to South Australian waters.
7. To develop methodologies for quality assurance with national applicability to the oyster farming industry.

Need

Australia currently supplies over 40% of the world's wild-harvested abalone and is also becoming increasingly involved in development of the abalone aquaculture industry. As wild-harvested abalone stocks decline demand for aquacultured abalone will increase and prices will continue to rise. Most commercial abalone can grow 70-80mm in 3-4 years. Although slow, growth rates have been significantly improved by considerable efforts from researchers and industry, particularly in manufactured diets and improved tank technology. As abalone are a slow growing species with enormous commercial potential for Australia, it is crucial to develop a sensitive and reproducible method for the measurement of growth rates and to be able to study the effects of various husbandry practices and diets on abalone development. Thus any gains made in reducing grow-out time will have a significant economic impact on this developing industry.

The identification and production of MIPs and other novel insulin-like proteins (ILPs) in abalone would allow studies to be performed to examine their roles in growth and development. This in turn could have major implications for the abalone aquaculture industry. The development of endocrinological assays as indicators of growth potential or response to factors such as diet could provide valuable tools for maximising the output of abalone farms in the most cost-efficient manner. This project addresses production, the highest ranked Research and Development priority for the aquaculture sector identified in the S.A. Fisheries and Aquaculture Five Year Research and Development Strategy. This area has been recognised as vital for the management of a viable abalone aquaculture industry and the study we propose has direct implications for improving growth and marketing characteristics in abalone.

Objectives

1. To use molecular biology techniques to isolate insulin-related peptides (ILPs) from abalone.
2. To use these peptides to produce reagents and develop immunoassays for measuring IRP concentrations in abalone.
3. To characterize the expression of these factors at the DNA and protein levels.
4. To undertake a preliminary study examining the correlation of the ILP levels with growth.

Breeding F1 hybrid oysters from doubled haploids

Need

A major problem facing Pacific oyster growers throughout southern Australia is maintaining consistent product quality. High levels of genetic variability in oysters cause considerable variation in several economically important traits such as growth rate and condition. Variation in growth rate causes extra work because of the need for frequent grading, while variation in condition within a batch may prevent growers from obtaining a market for their oysters. Even for those oysters which are of sufficient quality to reach the market, supplying oysters that vary in condition, or colour for example, creates a perception of poor quality control and undermines market confidence in the suppliers (and hence in the growers). This variability in product quality is currently unavoidable simply because of the intrinsic genetic variation in oyster stocks.

If genetically uniform oysters could be produced, then a major source of variation in product quality would have been removed from the oyster industry. All oysters grown from a batch of genetically identical spat should respond similarly in any given environment. Consequently, considerable improvements in consistency of product quality and appearance could be achieved. These genetically uniform oysters would still show some variation in (e.g.) growth rate and degree of condition due to differences in the environments they experience — for example, oysters which had been growing at the centre of a basket or lease may gain condition more slowly than those at the edge — however, because this variation would now be solely due to environmental factors, growers would be able to rapidly identify and control these factors. This would increase consistency of product quality still further.

A second major limitation to the oyster industry at present is a lack of stock which have been genetically selected to produce commercially desirable characteristics. Given the wide variation in environmental conditions under which Pacific oysters are grown in Australia (from cold, low-salinity waters off Tasmania, to warm, high salinity waters in S.Australia) considerable opportunity exists to breed oysters which have been genetically selected to perform well in a given environment. The Pacific oyster industry has recently engaged in a research programme which aims to selectively breed oysters that match grower and market demands more closely than present stocks. However, this programme uses conventional breeding techniques, and (commercial) benefits to growers cannot be expected for some considerable time.

Objectives

1. To obtain at least 20 (and preferably 100) families of doubled haploid Pacific oysters
2. From some of these doubled haploid oysters, establish inbred lines that contain both sexes so that they can be maintained by normal breeding procedures
3. To use these inbred lines to trial a range of F1 hybrids and identify the best F1 hybrid (or hybrids) for commercial production of genetically uniform batches of oysters

Objectives

1. To compare biochemical methods of assessing the effects of pollutant stress in tuna with methods currently used. New methods include measurement of adenylates and ATPase in fish tissue samples.
2. To evaluate methods of sampling fish or fish tissues from pens which will reduce stress and improve the probability of recovery of sampled fish which are returned to the pens. This part of the project will be closely co-ordinated with workers evaluating current sampling practices.
3. To evaluate the use of the biochemical responses of other species (e.g. tommy ruff, blue mussels) which cohabit with tuna in farm pens as surrogate indicators of pollutant stress experienced by tuna.
